چکیده

1 21 We examined the relative amounts of Na –Ca exchanger (NCX) isoform mRNAs in cultured neurons, astrocytes and developmental rat brain. NCX1 transcript was predominant in neurons and astrocytes, but NCX2 transcript was about four-fold higher than NCX1 or NCX3 transcript in adult rat cortex. NCX2 transcript in the cortex increased markedly during postnatal development, whereas NCX1 and 1 45 21 NCX3 transcripts decreased. Na -dependent Ca uptake in the cortical homogenate increased significantly during postnatal development.  2000 Elsevier Science B.V. All rights reserved. Developing Rabbit and Rat Hearts

To functionally compensate for an underdeveloped sarcoplasmic reticulum in immature cardiomyocytes, it has been proposed that the sarcolemmal Na+-Ca2' exchanger may assume a more predominant role for regulating cytosolic Ca2`. Previous studies using sarcolemma prepared from developing rabbit hearts demonstrated that Na+-dependent Ca2` uptake and exchanger protein content were highest at birth a...

Colocalization of voltage-gated Na+ channels with the Na+/Ca2+ exchanger in rabbit cardiomyocytes during development.

Reverse-mode activity of the Na(+)/Ca(2+) exchanger (NCX) has been previously shown to play a prominent role in excitation-contraction coupling in the neonatal rabbit heart, where we have proposed that a restricted subsarcolemmal domain allows a Na(+) current to cause an elevation in the Na(+) concentration sufficiently large to bring Ca(2+) into the myocyte through reverse-mode NCX. In the pre...

Exchanger from Drosophila melanogaster

Ion transport and regulation were studied in two, alternatively spliced isoforms of the Na 1 -Ca 2 1 exchanger from Drosophila melanogaster . These exchangers, designated CALX1.1 and CALX1.2, differ by five amino acids in a region where alternative splicing also occurs in the mammalian Na 1 -Ca 2 1 exchanger, NCX1. The CALX isoforms were expressed in Xenopus laevis oocytes and characterized ele...

Pharmacological inhibition of the Na(+)/Ca(2+) exchanger enhances depolarizations induced by oxygen/glucose deprivation but not responses to excitatory amino acids in rat striatal neurons.

BACKGROUND AND PURPOSE Neuronal Na(+)/Ca(2+) exchanger plays a relevant role in maintaining intracellular Ca(2+) and Na(+) levels under physiological and pathological conditions. However, the role of this exchanger in excitotoxicity and ischemia-induced neuronal injury is still controversial and has never been studied in the same neuronal subtypes. METHODS We investigated the effects of bepri...
